Isoflavones Quantification by Capillary Electrophoresis in Soybean Seeds: Effects of Variety and Environment

Soybean isoflavones (genistin, daidzin, glycitin, the malonyl form and their aglucons) are supposed to be responsible for soyfoods astringency. Generally, High Performance Liquid Chromatography with a methanol/water elution gradient, is used for isoflavone quantification. Recently, Capillary Zone Electrophoresis (CZE) has been used to separate several flavonoids in plant extracts. This report presents the results of CZE for the analysis of isoflavones in soybean seed extracts. CZE separation conditions were optimized for soybean isoflavones.

Different extraction conditions (modification of temperature and solvent) were compared using CZE. Extraction temperature did not affect total isoflavones content but the composition. Indeed, malonyl form were thermally instable.

Isoflavones content of seeds from different varieties grown on different sowing dates were analyzed. Total isoflavone contents showed an important variability among varieties and between sowing dates. Variety - sowing date interactions also affected isoflavone composition.
The choice of variety and environment conditions can contribute to improve seed quality by reducing isoflavones content and/or by modifying their composition.
