EFFECTS OF ISOFLAVONE AND FLAVONOIDS ON QUAIL SKELETAL MUSCLE CELL GROWTH IN VITRO

Skeletal muscle cells from 8-d-old qualil embryo were cultured with medium (MEM) free of serum and embryo extracts for 48 hrs (from 24 hrs to 72 hrs after plating) to study the effects of four levels (10-9, 10-8, 10-7 and 10-6 mol/L, n=8) if daidzein, formononetin, quercetin and leuteolin on the concentrations of creatine phosphokinase (CPK) and total proteins (TP) in muscle cell cultures. It was found that isoflavonic phytoestrogen, daidzein and fomononetin (10-9 ti 10-6 mol/L) had no obvious effect on skeletal muscle cell growth (P>0.05). Of flavonoids, quercetin of 10-7 and 10-6 mol/L exhibited stimulating effects, 66.5% (P<0.01) and 194% (P<0.01) increase in CPK, 13.0% (P>0.05) and 21.0% (P<0.05) increase in TP, whereas leuteolin of 10-6 mol/L showed inhibitory effects, 18.6% (P<0.05) decrease in CPK and 8.7% (P>0.05) decrease in TP. It indicated that the isoflavonic phytoestrogen (daidzein and formonetin) has no effects on quail skeletal muscle cell growth in vitro, however flavonoids quercetin with Beta-adrenergic effects exhibited promoting effects.
